Something to be aware off if you use the Lightning Conductor 2010 Web Part and disable My Sites and the User Profile Service Application. This causes the LCWP to return an error‘No User Profile Service is available in the current context’.
A workaround is to configure the User Profile Service Application service and remove permissions to My Sites (found at Central Administration > Application Management > Manage service applications > User Profile Service Application > Manage > Manage User Permissions)
